I hope they give this one to Ramis, but I'm more worried about the script and studio rewrites (ie: this needs to sell more toys, and we don't think the audience will get your jokes) than anything else.
What do you have against Ghostbuster II ? While not on par with the first, it's still very, very good. And while I'll admit that My Super Ex-Girlfriend was stupid, Evolution was an awesome movie.
I don't see why you would be agaisnt the idea of the original director returning? While Ramis is a good choice too, I wouldn't be against Reitman. After all, he made the ifrst two what they are now...
@Evil Tortie's Mom: R.O.A.C.H.: Yes! It would seriously not be right without Sigourney, and I am so, so hoping that the direction will fall to somebody, anybody good. Ramis would be great, but I'd be perfectly happy with any competent director (i.e., not Reitman).
@Mathmos: I think Anna Faris is the best choice. She's consistently good to great, despite being in some less than good movies. She always fully commits to her roles no matter how terrible the character is, and has shown to be fine for slapstick humor.
Amanda Bynes may not be as "hot" as some of the other choices, but she's at the very least attractive. She's pretty adept at physical comedy as well.
I think Rashida Jones would be great too, but I think she'd be great in just about anything.

I'd even suggest Claudia Black, Vala Mal Doran was almost my only reason for watching the final two seasons of Stargate SG-1, and I think he handled her duties as comic relief quite well.

@RRich: Ever since watching her in Low Down Dirty Shame, I have had a little crush on her. I agree that she would be able to pull the roll off perfectly.
@Alasdair5000: Tina Fey's only problem is that at this point she might be reaching critical overload in terms of over-exposure. Otherwise, I could support that casting decision.
Eliza Dushku? Oh god, no. I know it's going to piss of the Whedonites, but she cannot act, let alone be genuinely funny without seeming horribly forced. If Ghostbusters 3 is ever to become a reality (and I refuse to believe it until I see a trailer at my local theater - my heart's been broken too many times before), there should certainly be a female recruit, but I'd prefer her to be played by Kristen Wiig. She's got a wicked sense of comic timing and I think would ideally gel with the core foursome's brand of dry banter.
@Arceus: You're flinging "amazing" around with careless abandon. And a 2 point deduction for "uncanny". The only things Eliza Dushku can do that are amazing and/or uncanny is stand around being hot, and while that's awesome it's certainly not Ghostbuster material.
I've never seen anything from Wiig that would lead me to believe that she is capable of dry or nuanced humor at all. If she works on SNL, and I think it is hit or miss, it is because of the nature of sketch comedy where it is ok for a character to have obnoxious ticks/mug for the camera without any justification.
